Emergency services were called to Byrnes Road near Bavin Road, between North Wagga and Bomen, around 11.30am.
Paramedics were responding to reports of a truck on its side and on fire, and a driver with minor injuries, a spokesman said.
Fire and Rescue NSW crews raced to the scene to tackle the blaze, with support from NSW Rural Fire Service in tackling the fire.
The sheep truck was believed to be heading to the Wagga Livestock Marketing Centre before the accident.
A Local Land Services (LLS) spokesperson confirmed that LLS staff were at the scene assisting.
It is understood traffic is affected at the Byrnes Road roundabout, with motorists being diverted away from the area. North Wagga's Hale Street has also been closed off.
This is the third livestock truck roll over in the Riverina since February 11, when more than 20 steers were euthanised after a B-double rolled on the Hume Highway near Tarcutta. A week later, a number of sheep had to be put down after another B-double rolled near Collingullie.
